Experimental evaluation of super-resolution imaging and magnification choice in single-particle cryo-EM

The resolution of cryo-EM reconstructions is fundamentally limited by the Nyquist frequency, which half sampling frequency detector and depends upon magnification used. In principle, super-resolution imaging should enable to surpass physical limit increasing yet there are few reports that do so. Here we directly examine contribution information, obtained with K3 direct electron using a 2-condenser microscope, single-particle surpassing limit. We also present comparative analysis sample imaged at four different magnifications. This demonstrates lower magnifications can be beneficial, despite loss higher signal, due increased number particle images obtained. To highlight potential utility data collection, produced 3.5 Å reconstruction jack bean urease particles from single micrograph.
